胚胎中晚期皮肤微小RNA差异表达谱及生物学功能分析
Differentially expressed miRNAs profiles in mid- and late-gestational fetal skin and biological functions
摘要目的 通过比较胚胎中晚期皮肤中微小RNA(miRNA)表达谱差异,探讨miRNA在皮肤表型转变中的生物学功能.方法 应用芯片高通量筛选出3组胚胎第16和19天皮肤样品中差异表达的miRNA分子.miR-29b/c分子的表达差异经定量逆转录-聚合酶链反应(RT-PCR)验证.应用生物信息学软件miRGator和DAVID对差异MiRNA的靶基因进行预测和生物学功能分析.结果 E16和E19皮肤中共有16个差异表达的miRNA分子.定量PCR结果显示miR-29b/c,分别为26.4和16.7倍差异.所预测的1295个靶基因可能参与了多种信号通路包括转化生长因子(TGF)-β通路.结论 胚胎中晚期皮肤中差异表达的miRNA可能参与了胚胎皮肤表型的转变且在无瘢痕愈合过程中发挥重要作用.

abstractsObjective To identify differentially expressed microRNAs (miRNAs) between midand late-gestational fetal skin and elucidate the possible roles of miRNAs involved in the fetal skin phenotypic transition.Methods Comprehensive miRNA profiling of E16 and E19 mouse skin was performed by miRNA microarray.The expression levels of miR-29b/c were further confirmed by real-time reverse transcription-polymerase chain reaction (RT-PCR).Putative target genes of miRNAs were predicted by computer algorithms MiRGator and functionally annotated by DAVID bioinformatics resources.Results Sixteen miRNAs were identified to be differentially expressed between E16 and E19 fetal skin.The expression levels of miR-29b and miR-29c determined by real time RT-PCR were 26.4-fold and 16.7-fold,respectively.Bioinformatics analysis of 1295 putative targets estimated by computational programs showed that theses target genes were potentially involved in the several signal pathways including TGF-β pathway.Conclusion Our findings have revealed several differentially expressed miRNAs between mid- and late-gestational fetal skin,suggesting that miRNAs might contribute to fetal skin phenotypic transition and specific miRNA might probably be implicated in the scarless wound healing.
