双色荧光示踪裸小鼠模型用于脑胶质瘤组织重构的研究

摘要目的 探讨双色荧光示踪裸小鼠模型用于脑胶质瘤组织重构研究的实用价值.方法 将转染红色荧光蛋白基因(RFP)的SU3、U87-MG和用亲脂性碳青染料(CM-DiI)染色的C6胶质瘤细胞6.7×109/L,借助立体定向仪接种于自家培育的表达绿色荧光蛋白(GFP)近交系裸小鼠脑内,建立RFP/GFP双色荧光示踪的肿瘤模型,分析示踪效果.结果 SU3、U87-MG和C6转染或染色成功的比例接近100%,这些细胞原位移植成功率为100%(30/30);移植瘤组织重构是在侵袭于脑实质、脑室、脑血管的肿瘤细胞诱导或与宿主细胞融合后发生的.结论 在肿瘤组织重构研究中,双色荧光示踪模型对区别肿瘤细胞与宿主细胞之间发生的组织细胞学变化起独特作用.

abstractsObjective To explore the practical value of dual-color fluorescent tracing nude mouse model used in the study on glioma tissue remodeling.Methods SU3,U87-MG transfected with the red fluorescence protein (RFP) gene and C6 traced with lipophilic carbon blue dye (CM-DiI),6.7 × 109/L,were injected slowly to the caudate nucleus of the enhanced green fluorescent protein (EGFP) nu/nu mice with the help of the stereotactic instrument and micro-injector,a dual-color fluorescent tracing model was established and the effect of tracer was analyzed.Results (1) SU3,U87 and C6 almost 100% expressed red fluorescence under the fluorescence microscope,and orthotopic transplant success rate was 100% (30/30) ; (2) The occurrence of tumor tissue remodeling was induced by tumor cells and/or after fusing with host cells.Conclusion Dual-color fluorescent tracer model may play a unique role in distinguishing the histological changes between tumor cells and host cells in the study on tumor tissue remodeling.
