摘要目的 探讨肺癌、癌旁及正常组织中微小RNA(miRNAs,miR)表达谱差异及其与TNM病理分期的关系.方法 采用Agilent miRNA芯片检测9例非小细胞肺癌患者的同体癌、癌旁和正常组织的miRNA表达谱,比较不同TNM分期间的表达差异.结果 筛选出36个在癌组织和癌旁组织之间差异表达的miRNAs和55个在癌组织和正常组织间差异表达的miRNAs;与早期患者比较,Ⅲa期的正常组织有hsa-miR-205、hsa-miR-34b等11个差异表达的miRNAs;而癌旁组织则从Ⅱa期开始就出现了9个差异表达的miRNAs;不同TNM分期的癌组织之间仅有hsv2-miR-H7-3p和hsa-miR-23a*表达差异有统计学意义(P＜0.05).结论 随着肺癌病理分级的发展,癌旁组织甚至远端组织也会出现特定mircoRNA表达量的变化,该特定mircoRNA的表达可能提示肿瘤的浸润能力.

abstractsObjective To study the difference spectrum of microRNAs (miRNAs,miR) expressions among lung cancer tissues,adjacent tissues and normal tissues and its correlation with the Tumor,Node and Metastasis (TNM) staging.Methods Agilent miRNA expression microarray was used to detect nine cases of non-small cell lung cancer,adjacent tissues and normal tissues of the distal end.And then compare the expression profiles between different stages.Results We screened out 36 differentially expressed miRNAs between adjacent tissues and cancer tissues,and 55 differentially expressed miRNAs between cancer tissues and normal tissues.Compared with distal normal tissues of patients with early stage lung cancer,distal normal tissues of patients with Ⅲ a staging have 11 differentially expressed miRNAs,including hsa-miR-205,hsa-miR-34b,et al.However,according to adjacent tissues,11 differentially expressed miRNAs were screened between patients with Ⅰ a and Ⅱ a staging.According to cancer tissues,hsv2-miR-H7-3p and hsa-miR-23a * were differentially expressed among Ⅰ a,Ⅱ a and Ⅲ a stagings.Conclusion With the increasing of TNM staging,adjacent tissues and even distal tissues also occur on specific mircoRNA expression changes,which maybe suggest the ability of tumor invasion.
