半乳糖凝集素-3和67000层黏连蛋白受体蛋白和mRNA在四肢静脉血管畸形组织中的表达及意义
Detection of galectin-3 and 67 000 laminin receptor protein and mRNA in venous malformation of extremities
摘要目的 探讨半乳糖凝集素-3(Galectin-3)和67000层黏连蛋白受体(67LR)蛋白和mRNA在四肢静脉血管畸形组织中的表达及意义.方法 采用免疫组织化学及原位杂交方法检测165例四肢静脉畸形组织和20例远端手术切缘正常组织中Galectin-3和67LR蛋白及其mRNA的表达,并分析其与四肢静脉血管畸形患者临床病理特征之间的关系.结果 四肢静脉血管畸形组织中Galectin-3和67LR阳性率分别为83% (138/165)、76%(126/165),高于正常组织中的15% (3/20),10%(2/20,P<0.05);其两者mRNA阳性率分别为86% (143/165)、78%(129/165)高于对照组的20% (4/20),15% (3/20,P<0.05).四肢静脉血管畸形组织中Galectin-3和67LR蛋白及其mRNA的阳性表达均与患者的年龄、性别无明显相关(P>0.05);而Galectin-3和67LR蛋白及其mRNA的阳性表达率,在局部侵袭深度深者明显高于侵袭深度浅者(x2=22.051、5.525,P<0.05),且两者的阳性表达率在无明显界限者中明显高于界限清者(x2=7.756、9.599,P<0.05).结论 四肢静脉血管畸形组织中Galectin-3和67LR蛋白及其基因mRNA的表达可能在基因转录水平已经上调,2个基因可能参与促进了静脉血管畸形的发生、发展,与静脉血管畸形在四肢局部侵袭破坏密切相关.

abstractsObjective To study the role of galectin-3 and 67 000 laminin receptor (67LR) protein and mRNA expression in venous malformation of extremities tissue.Methods The expressions of galectin-3 and 67LR protein and mRNA were detected using by immunohistochemical staining and in situ hybridization in 165 cases of venous malformation of extremities.Results The positive rate of galectin-3 and 67LR and protein were 83% (138/165) and 76% (126/165) in venous malformation of extremities tissue.The positive rate of galectin-3 and 67LR and mRNA were 86% (143/165) and 78% (129/165) in venous malformation of extremities respectively,whereas the epithelial cells of extremities tissue from the distant cutting margin had weak reactivity which were 15% (3/20) and 10% (2/20),20% (4/20) and 15% (3/20) respectively (P <0.05).The expression of 67LR and galectin-3 protein and mRNA in venous malformation of extremities tissue were not related to gender and age (P > 0.05).A significant association between the expression of galectin-3,67LR mRNA and depth of venous malformation invasion was found (x2 =22.051,5.525,P < 0.05).And enhanced expression of galectin-3 and 67LR mRNA was found in the without clear boundary tissue (x2 =7.756,9.599,P < 0.05).Conclusion Galectin-3 and 67LR protein and mRNA expressions are higher in venous malformation of extremities tissue,two genes may be involved in promoting the occurrence and development of the vein abnormalities,and venous vascular malformation in the limb is closely related to the local attack damage.
