Construction of three-dimensional culture model of human neuroendocrine tumor BON-1 and H727 cell lines

摘要目的 建立人神经内分泌瘤细胞的体外三维培养模型.方法 制备琼脂糖凝胶、海藻酸盐/聚丙烯酰胺共聚物(ALG/PAAm)水凝胶及聚乙二醇(PEG)交联聚甲基乙烯基醚-alt-马来酸(PMVE-alt-MA)水凝胶,基于这3种材料构建人神经内分泌瘤H727及BON-1细胞株三维培养模型,每24h通过倒置相差显微镜观察其形态学变化.结果 三维培养下H727及BON-1细胞的形态不同于二维平面培养;培养至1周左右,H727细胞在琼脂糖凝胶、ALG/PAAm水凝胶及PEG交联PMVE-alt-MA水凝胶中分别形成了球径350~ 450、150 ~ 250及50~100μnn的多细胞球体;而BON-1细胞在PEG交联PMVE-alt-MA水凝胶中则生长成球径150 ~ 200 μm的多细胞球体,但在另外两种材料中仅形成形状不规则的多细胞聚集体.结论 该3种材料可应用于H727及BON-1细胞株体外三维培养模型构建,且各有优劣之处,可根据具体的实验要求选择合适的培养材料.

abstractsObjective To provide new methods for the study of human neuroendocrine tumor cells in vitro by establishing three-dimensional culture models.Methods Agarose gel,alginate/polyacrylamide copolymer (ALG/PAAm) hydrogel and polyethylene glycol (PEG) crosslinked poly methyl vinyl ether maleic acid alt (PMVE-alt-MA) hydrogel were prepared.Three-dimensional culture models of human neuroendocrine tumor H727 and BON-1 cell lines based on these three materials were constructed,and the morphological changes of the models were observed by a microscope every 24 h.Results The shapes of H727 and BON-1 cells with three-dimensional cuhure were different from those of two-dimensional culture.Mter cultured for one week,H727 cells formed different multicellular spheroids with sphere diameter of 350-450,150-250 and 50-100 μm on agarose gel,ALG/PAAm hydrogel and in PEG crosslinking PMVE-alt-MA hydrogel respectively.And BON-1 cells grew up into muhicellular spheroids with sphere diameter of 150-200 μm in PEG crosslinking PMVE-alt-MA hydrogel,but they just formed irregularly shaped multicellular aggregates on the other two culture materials.Conclusion The three materials could be applied for constructing three-dimensional culture models of BON-1 and H727 cell lines.Due to their advantages and disadvantages,the appropriate culture material would be selected according to specific experimental requirements.
