四肢静脉血管畸形组织中67000层粘连蛋白受体基因的检测及其临床意义
The clinical meaning and measurement of 67 000 laminin receptor in extremities venous malformation tissue
摘要目的 检测67000层粘连蛋白受体(67LR)基因在四肢静脉血管畸形组织中的表达,探讨67LR基因与四肢静脉血管畸形发生和进展的关系.方法 收集12例新鲜四肢静脉畸形组织标本,单纯性大隐静脉曲张组织标本12例做为对照组,采用Western blot及实时荧光定量聚合酶链反应(Real-time PCR)检测67LR的表达.结果 静脉畸形组和对照组67LR蛋白及mRNA的相对表达量分别为0.859±0.388、0.219±0.202和5.31×10-3±8.82×10-4、(4.93±5.41)×10-4;眼部静脉畸形组织中半乳糖凝集素-3在mRNA及蛋白水平上的表达量明显高于对照组(P=0.041,0.019),并随着病变侵及深度及范围的加重有递增的趋势.结论 67LR基因在四肢受累严重的组织中表达高于轻度病变或表浅者,其表达异常与静脉畸形局部侵袭发展关系密切,对该基因表达异常的检测,可用于判断局部侵犯程度.其四肢静脉血管畸形的发病机制可能与67LR基因的参与密切相关.

abstractsObjective The development of extremities venous malformation (VM) severely impaired extremities shape and functions.Investigations showed that 67 000 laminin receptor (67LR) was closely correlated with local invasion of extremities VM, further indicating that 67LR played a pivotal role in the mechanisms underlying ocular VM development.To measure the expression level of 67LR in the extremities VM tissue, and explore its associations with the occurrence and development of extremities VM.Methods Collect 12 fresh extremities VM tissue samples, and 12 simplex great saphenous vein varices tissue samples as control.Using Western blotting and real-time quantitative polymerase chain reaction (Real-time PCR) to measure 67LR expression levels.Results The relative mRNA and protein levels of 67LR in VM and control group were 0.859±0.388, 0.219±0.202 and 5.31×10-3±8.82×10-4, (4.93±5.41)×10-4, respectively;67LR levels in both mRNA and protein in extremities VM tissue were obviously higher than control group (P=0.041, 0.019), an increasing trend was companied with aggravating pathological changes in both degree and range.Conclusion 67LR was highly expressed in extremities VM tissue in both mRNA and protein levels, it might closely correlate with ocular VM development and prognosis, could be used to determine the degree of local invasion of extremities VM.The pathological mechanism of extremities VM may associate with 67LR involvement, will be further investigated using genetic chip.
