环状RNA CircPGC在胃癌组织中低表达及其促进胃癌细胞迁移
Circular RNA CircPGC expression level is decreased in gastric cancer tissues and it promotes migration of gastric cancer cells
摘要目的 探讨CircPGC在胃癌中的表达及其对胃癌细胞AGS迁移的调控作用.方法 通过实时定量反转录聚合酶链反应(RT-qPCR)检测CircPGC表达水平;无缝克隆法构建CircPGC过表达载体pcDNA3.1(+)-CircPGC;划痕实验检测CircPGC对AGS细胞迁移的影响;应用GraphPadPrism 6.0统计软件分析,计量资料以均值±标准差(Mean±SD)表示,组间比较采用t检验.结果 人胃癌组织中CircPGC的表达水平(△Ct=12.480±2.385)显著低于胃正常组织(△Ct=7.327±2.696)和癌旁组织(ACt=7.122±3.030).与对照组比较,转染pcDNA3.1(+)-CircPGC质粒后AGS细胞中CircPGC表达量明显增高(t=11.860,P<0.01),过表达CircPGC促进细胞迁移:划痕实验结果显示,过表达组细胞迁移距离(44.900 ±5.284)%较对照组(10.010±1.597)%长(t=10.580,P<0.01).结论 CircPGC是与胃癌迁移相关的环状RNA.

abstractsObjective To investigate the expression of CircPGC in gastric cancer and regulation on the migration of gastric cancer cell AGS.Methods Real-time quantitative reverse transcriptase-polymerase chain reaction (RT-qPCR) was used to verify the level of CircPGC.The CircPGC overexpression vector,pcDNA3.1 (+)-CircPGC,was constructed by seamless cloning method.The effect of CircPGC in migration of AGS cell was detected by scratch assay.Using GraphPad Prism 6.0 statistical software analysis,the measurement data were expressed as mean ± standard deviation (Mean ± SD),and t-test was used for comparison between groups.Results The expression level of CircPGC in human gastric cancertissues (△Ct =12.480 ± 2.385) was significantly lower than that in normal gastric tissues (△Ct =7.327 ±2.696) and adjacent tissues (△Ct =7.122 ±3.030).Compared with the control group,the expression of CircPGC in AGS cell was significantly increased after transfection of pcDNA3.1 (+)-CircPGC plasmid (t =11.860,P < 0.01).Overexpression of CircPGC promotes cell migration:The scratch test showed that the migration distance of AGS cells in the overexpressing group was (44.900 ± 5.284)%,which was longer than the control group (10.010 ± 1.597) % (t=10.580,P<0.01).Conclusion CircPGC is a circular RNA associated with migration of gastric cancer.
