腺性肥大乳房乳腺组织和上皮细胞中雌激素受体α的定位和表达
The location and expression of estrogen receptor α in mammary tissue and epithelial cells of gland-associated hypertrophic breast
摘要目的:观察雌激素受体α(ERα)在腺性肥大乳房乳腺组织和上皮细胞中的定位和表达,探讨其在腺性乳房肥大症病因学上的意义。方法:对10例腺性乳房肥大症患者和正常体积乳房乳腺上皮细胞进行原代培养,采用免疫组织化学法(IHC)和免疫荧光组织化学法(IFHC)对乳腺组织的病理学特点进行观察,采用蛋白质印迹法(Western blot)检测组织及细胞中ERα蛋白的表达量。组间比较采用单因素方差分析。结果:ERα表达于导管和小叶的腔上皮细胞内,在肌上皮细胞和间质细胞等细胞内几乎不表达,且腺性肥大乳房乳腺组织导管上皮处ERα阳性细胞数多于小叶,两者比例较为恒定(1.7∶1.0)。ERα阳性细胞在腺性肥大乳房和正常体积乳房乳腺组织中的阳性表达率(PER)分别为17.25%和6.13%。腺性肥大乳房组、正常体积乳房组细胞株中的ERα蛋白表达值(PEV)为1.965和1.002。腺性肥大乳房乳腺组织中ERα PER和ERα PEV与正常体积乳房组间差异有统计学意义( P<0.05),且PER和ERα PEV与乳房肥大的严重程度呈正相关。 结论:ERα在乳腺组织内的表达定位具有特征性。与正常体积乳房乳腺上皮细胞比较,腺性肥大乳房乳腺上皮细胞的ERα PER和ERα PEV均明显增高,且ERα表达情况的高低与乳房肥大的严重程度成正相关。由此推测,腺性乳房肥大症的发生与乳腺组织内ERα的分布特点和上皮细胞内ERα表达增多有密切关。

abstractsObjective:To investigate the location and expression of estrogen receptor α (ERα) in mammary tissue and epithelial cells of gland-associated hypertrophic breast, and analyze their signification in etiology of gland-associated hypertrophic breast.Methods:To culture primary cells in vitro of hMEC-GAHB and human mammary epithelial cells from normal volume breast and to analyze qualitatively and quantitatively the histopathological characteristics of mammary tissue and the expression of ERα in tissue and cells by immunohistochemistry (IHC) and immunofluorescence histochemistry (IFHC) and Western blotting. Results:ERα expressed in the luminal cells of ducts and lobules, and ERα+ cells, which expressed in the mammary tissue of gland-asoociated hypertrophic breast, were more in ducts than those in lobules with the constant ratio of 1.7∶1.0. However, it didn’t express in myoepithelial cells or mesenchymal cells at all. The positive expression rate (PER) of ERα+ cells in mammary tissue of gland-associated hypertrophic breast and normal volume breast were respectively 17.25% and 6.13%. The protein expression value (PEV) of ERα in hMEC-GAHB and hMEC-NVB were respectively 1.965 and 1.002. PER and ERα PEV in gland-associated hypertrophic breast were both higher than those in normal volume breast, and they had a positive correlation with the severity of hypertrophic breast.Conclusion:The expression of ERα had a characteristic location. Compared with normal volume breast, the PER and PEV of ERα in hMEC-GAHB increased obviously, and have a positive correlation with severity of hypertrophy in gland-associated hypertrophic breast. Therefore, we could infer that the happen of gland-associated macromastia has a very close relationship with the expression and location of ERα in mammary tissue and epithelium.
