三叶因子3在下肢静脉血管畸形组织的表达及临床意义
Expression and clinical significance of trefoil factor 3 in lower extremity venous malformations
摘要目的:检测三叶因子3(TFF3)在下肢静脉血管畸形组织的表达,探讨其与下肢静脉血管畸形发生、发展的关系。方法:选择2019年09月01日至2020年8月31日郑州大学第三附属医院血管瘤外科行手术切除的26例四肢静脉畸形患者进行研究,选取26例四肢静脉畸形组织为实验组,26例距病变切缘3cm的正常静脉组织为对照组,采用免疫组织化学染色检测TFF3蛋白的阳性表达率;从26例四肢静脉畸形患者中筛选出下肢静脉畸形病变患者共11例进行研究,术中切除的11例下肢静脉畸形组织作为实验组;另收集距病变切缘3 cm的11例正常静脉组织为对照组,采用实时荧光定量聚合酶链反应技术(Real-time PCR)和蛋白质印迹实验(Western blot)检测TFF3 mRNA及蛋白的相对表达水平。计量资料采用 t检验,计数资料 χ2检验进行分析。 结果:免疫组化结果显示实验组中TFF3蛋白的阳性表达率为[80.77%(21/26)]明显高于对照组[11.54%(3/26)],差异有统计学意义( χ2=25.07, P<0.01);Real-time PCR结果显示实验组中TFF3 mRNA相对表达量(7.436±6.612)高于对照组(1.494±2.572),差异有统计学意义( t=2.778, P<0.05)。Western blot结果显示在实验组中TFF3蛋白的相对表达量(0.425±0.481)明显高于对照组(0.089±0.051),差异有统计学意义( t=2.304, P<0.05)。 结论:TFF3可能与下肢静脉畸形的发展和局部侵袭性有关。

abstractsObjective:To detect the expression of trefoil factor 3 (TFF3) in lower extremity venous malformation (LEVM) tissue, and to explore its relationship with the occurrence and development of LEVM.Methods:Totally, 26 paraffin specimens of LEVM confirmed by hematoxylin and eosin (HE) staining were selected as the experimental group, and 16 paraffin specimens of normal appendix tissues were selected as the control group. Immunohistochemical staining was used to detect the positive expression rate of TFF3 protein in venous malformations and normal appendix vessels. A total of 11 cases screened from 24 cases of LEVM were studied. The resected tissues of 11 cases of LEVM served as experimental group, and 11 cases of normal venous tissues 3 cm away from the resection edge of the lesions as the control group. The real-time fluorescent quantitative polymerase chain reaction (real-time PCR) and Western blotting were used to detect the relative expression levels of TFF3 mRNA and protein in the experimental group and the control group, respectively.Results:The results of immunohistochemistry showed that the positive expression rate of TFF3 protein in LEVM tissues [80.77% (21/26)] was significantly higher than that in normal venous tissues of appendix [18.75% (3/16), χ2=13.53, P<0.01]. Real-time PCR and Western blotting results showed that the relative expression levels of TFF3 mRNA and protein in LEVM tissues were 7.436±6.612 and 0.4250.481, respectively, which were significantly higher than in the control group (1.494±2.572 and 0.089±0.051, t=2.304, P<0.05). Conclusion:The relative expression of TFF3 mRNA and protein in LEVM increased significantly, which may be involved in the local invasion of LEVMs.
