微小RNA-146a在结肠癌组织的表达及其临床意义
Relationship between expression of microRNA-146-a and pathological features in colon cancer
摘要目的:探讨结肠癌组织中以及癌细胞株中的微小RNA(miR)-146a的表达并分析与病理特征之间的关系。方法:收集整理2017年10月至2018年11月广东医科大学附属医院42例结肠癌组织与正常结肠组织(距癌组织边缘>5 cm),采用实时荧光定量聚合酶链反应(RT-qPCR)检测miR-146a在结肠癌组织及正常组织中的表达,同时分析其与病理特征之间的关系,总结抑制miR-146a表达对结肠癌细胞凋亡、增殖等影响。采用方差齐性检验和 χ2检验进行统计学分析。 结果:通过检测发现42例结肠癌组织miR-146a相对表达量显著高于正常癌旁组织,低分化癌细胞lovo中miR-146a表达量均高于中、高分化癌细胞( F=11.80, P<0.01);miR-146a的高表达与患者结肠癌分化程度差、肿瘤过大、淋巴结远处转移等有关( t=10.23、6.78、8.55, P<0.01)。通过抑制miR-146a表达可以有效预防癌细胞增殖及克隆形成。 结论:检测miR-146a在结肠癌组织的高度表达可有助于预测淋巴转移、肿瘤病变程度。

abstractsObjective:To investigate the expression of microRNA (miR)-146a in colon cancer tissues and cancer cell lines and analyze the relationship between miR-146a and pathological features.Methods:From October 2017 to November 2018, 42 cases of colon cancer tissue and normal colon tissue (> 5 cm from the edge of cancer tissue) were collected in Affiliated Hospital of Guangdong Medical University. Real-time fluorescence quantitative polymerase chain reaction (RT-qPCR) was used to detect the expression of miR-146a in colon cancer tissue and normal tissue. At the same time, the relationship between miR-146a expression and pathological features was analyzed, and the effects of miR-146a expression inhibition on apoptosis and proliferation of colon cancer cells were summarized. Homogeneity of variance test and χ2 test were used for statistical analysis. Results:The relative expression level of miR-146a in 42 colon cancer tissues was significantly higher than that in normal para-cancer tissues, and the expression level of miR-146a in lovo of poorly differentiated cancer cells was higher than that in medium and well differentiated cancer cells ( F=11.80, P<0.01). The high expression of miR-146a was associated with poor differentiation of colon cancer, large tumor, and distant lymph node metastasis ( t=10.23, 6.78, 8.55, P<0.01). Inhibition of miR-146a expression could effectively prevent the proliferation and cloning of cancer cells. Conclusion:The high expression of miR-146a in colon cancer tissue can be used as a gold indicator to effectively predict lymphatic metastasis and the degree of tumor lesion, which has certain reference value for clinical treatment.
