葡萄糖浓度对干细胞源性心肌细胞成熟影响的研究进展
The effect of glucose concentration on the maturation of stem cell-derived cardiomyocytes
摘要诱导干细胞向心肌细胞分化方法的出现为构建心血管疾病模型、药物测试和心肌再生治疗开辟了新的途径。然而,获得的干细胞源性心肌细胞的表型与成人心肌细胞有很大的差异,表现不成熟的特征,具体表现为自发收缩、形态不规则、肌节成分不同和代谢方式差异等等。这些特征可能对其临床应用产生不利影响。因此,制定提高干细胞源性心肌细胞成熟度的新策略至关重要。本综述回顾了干细胞源性心肌细胞与成人心肌细胞的主要表型差异,总结了改变培养基中葡萄糖浓度对干细胞源性心肌细胞成熟度的影响,以期为促进干细胞源性心肌细胞的成熟提供新的理论依据和研究思路。

abstractsThe emergence of methods to induce stem cell differentiation into cardiomyocytes has opened up new avenues for constructing cardiovascular disease models, drug testing, and myocardial regeneration therapy. However, the phenotype of the obtained stem cell-derived cardiomyocytes differs significantly from that of adult cardiomyocytes-specifically manifested by spontaneous contraction, irregular morphology, different sarcomere components, and differences in metabolic patterns. These features may have adverse effects on their clinical application. Therefore, it is crucial to develop new strategies to improve the maturity of stem cell-derived cardiomyocytes. This review reviews the main phenotypic differences between stem cell-derived cardiomyocytes and adult cardiomyocytes, with a focus on changes in the metabolic patterns of cardiomyocytes. It also summarizes the effects of changing glucose concentration in the culture medium on the maturation of stem cell-derived cardiomyocytes, in order to provide new ideas for promoting the maturation of stem cell-derived cardiomyocytes.
