人类膜性肾病可能的抗原成份及其免疫沉积物形成、定位的免疫电镜研究

摘要本文利用胶体金免疫电镜技术显示,在膜性肾病(MN)早期阶段,IgG不仅存在于上皮下沉积物(ID)还集中位于上皮细胞内溶酶体样结构中。表明在人MN形成过程中,肾小球上皮细胞,可能起着更为主动的、对特异抗体的内摄及清除作用。GP 160是位于人类肾小球上皮细胞和近曲小管刷毛缘的共同抗原成份。应用胶体金双标记检测,发现在IgG和GP160伴随存在于MN肾小球上皮细胞内和上皮下ID中。表明,GP 160可能为引起MN的抗原物质。

abstractsThere were a few reports in the literature on human renal pathology deal with the presence of tubular brush border antigen (RTE) in the glomerular aggregates of idopathic MN. These findings presume that RTE source antigens might play a role in the pathogenesis of some cases of human MN.For the purpose of making clear the pathogenesis of MN and researching into the possible antigen in human MN, two aspects of study have been done.The development of IDs in human MN was first demonstrated in this study. In early stage MN, IgG was located not only in dense deposits of subepithelium, surface of epithelium, but also accumulated in lysosome, as the same in PHN. It is suggested that in human being MN, GEC, as the main attacked target, may also play an active role in the endocytosing, transporting and clearing of IgG.Ten cases of MN and two cases normal renal tissue were studied prospectively by immunoperoxidase and immunoelectromicroscopic technique. The mAb Uro-II (anti-GP 160) staining revealed that there was a common antigen (GP160) among proximal tubules (PT) and GEC. Im munoelectromicroscopy double staining revealed that Uro-II (GP160) in company with IgG was located in the dense deposits of subepithelium and in the endosomes. It is suspected that GP 160 acts in the same way as GP 330 in HN, may play an antigenic role in the human MN.
