Effect of quinidine sulfate on the mRNA expression of Kv4.3 gene in neonatal rat ventricular myocytes
摘要目的 探讨硫酸奎尼丁对乳大鼠心室肌细胞Kv4.3基因mRNA水平表达的影响及其治疗Brugada综合征的分子机制.方法将乳大鼠心室肌细胞随机分为对照组及硫酸奎尼丁干预组,培养至24、48、72 h,应用半定量逆转录-聚合酶链反应(RT-PCR)技术检测Kv4.3基因mRNA表达的变化.结果 干预至24 h对Kv4.3基因mRNA水平表达无明显影响;至48 h,各干预组较对照组显著降低(P<0.05),且5与10 μmol/L组较1 μmol/L组下调作用更为显著(P<0.05);至72 h,各干预组较对照组显著降低(P<0.05),而各干预组间差异无统计学意义.结论 硫酸奎尼丁可显著下调Kv4.3基因mRNA水平表达,且较长时间干预时低浓度与较高浓度下调的幅度相近.

abstractsObjective To investigate the effect of quinidine sulfate on the mRNA expression of Kv4.3 gene of transient outward potassium channel in neonatal rat ventricular myocytes(NRVMs) and its possible an-tiarrhythmic mechanism of Brugada syndrome. Methods NRVMs were randomly divided into control group and intervention groups of different quinidine sulfate concentration, incubated to 24,48 and 72 h respectively. The mRNA expression of Kv4.3 gene was semi-quantified by reverse transcription-polymerase chain reaction. Results The differences of Kv4.3 gene expression among all groups did not reach statistical significance after interfered for 24 h. When incubated up to 48 h, the mRNA expression of Kv4.3 gene was significantly decreased in interfered groups compared with the control group (0.44,0.27,0.22 vs 0.54,P <0.05,respectively). Fur-thermore,middle and high dose of qunidine sulfate showed greater inhibition than the low dose. But after 72 h intervention,qunidine sulfate in all groups showed similar down-regulation effects on Kv4. 3 gene expression (0.25,0.21,0.21 vs 0.57, P < 0.05, respectively). Conclusion The mRNA expression of Kv4.3 gene is down-regulated after incubation with quinidine sulfate. In addition, with intervention for long time, the expression of Kv4.3 gene in the low dose is almost the same as in the high dose.
