多环芳烃暴露与外周血淋巴细胞核质桥和核芽的关系研究
Association between polycycle aromatic hydrocarbons exposure levels and nuccleoplasmic bridge and nuclear bud frequencies in coke-oven workers
摘要目的 寻找评价多环芳烃致外周血淋巴细胞遗传物质损伤的新的效应性生物标志物.方法 采集158名焦炉工和69名非多环芳烃暴露者肘静脉血1 ml,应用外周血淋巴细胞胞质分裂阻滞微核法制备涂片,计数核质桥和核芽发生率,评价外周血淋巴细胞的染色体损伤水平;收集研究对象的职业史、年龄、性别、吸烟和饮酒等信息.结果 焦炉工外周血淋巴细胞核质桥率为(9.41±3.73)‰,高于对照组的(1.88±1.49)‰,差异有统计学意义(t=-16.2,P<0.01);其核芽率为(7.13±4.01)‰,与对照组[(2.20 4-1.73)‰]相比差异有统计学意义(t=-9.79,P<0.001).发现核质桥和核芽率与多环芳烃暴露等级具有剂量.效应关系.与男性焦炉工相比,女性焦炉工具有较少的核质桥[(7.14 4-3.04)‰比(9.63±3.73)‰,t=-2.62,P=0.01]和核芽[(4.71±2.73)‰比(7.36±4.05)‰,t=-2.33,P=0.02]发生率.未发现年龄、吸烟和饮酒等对焦炉工的核质桥和核芽率的影响.结论 多环芳烃可致暴露工人染色体损伤增加,核质桥和核芽可以作为焦炉工染色体损伤的效应性生物标志物.
