初发型翼状胬肉患者组织中p53表达及DNA含量分析
Expression of p53 and flow cytometric analysis of DNA index and S-phase fraction in pterygium
摘要目的 研究初发型翼状胬肉组织p53的表达及DNA指数(DNA index,DI)和S期细胞比率(S-phase fraction,SPF),探讨以上因素在翼状胬肉发病中的作用.方法 免疫组织化学SP法对2005年8月至2006年12月间在南通大学附属医院眼科行手术治疗的84例(96眼)初发型翼状胬肉患者翼状胬肉组织进行p53表达检测,流式细胞术(flow cytometry,FCM)检测翼状胬肉组织细胞DNA指数(DNA index,DI)和s期细胞比率(S-phase fraction,SPF);并与32眼正常结膜组织进行对照.结果 p53在翼状胬肉的表达明显高于正常结膜(P<0.01);翼状胬肉和正常结膜组织相比,DI、SPF值均增高,但差异未达到统计学意义(P>0.05).结论 翼状胬肉组织P53的高表达及DI值、SPF值的轻度增高,提示翼状胬肉的发病机制可能是由于细胞增殖异常及凋亡失衡引起.

abstractsObjective To study the expression of p53 and DNA index(DI) and S-phase fraction (SPF) in pterygium, and explore their contribution of pathogenesis of pterygium. Methods Surgically excised pterygia of 84 cases (96 eyes) with newly developed pterygium and 30 samples of normal conjunctiva as control were collected for immunohistochemistry to detected the expression of p53. The DI value and the SPF value were detected in all specimens with FCM. Results The expression of p53 in pterygium was significantly higher than that of normal conjunctiva ( P < 0. 001). The DI value and the SPF value in pterygium were higher than that of normal conjunctiva but without statistical significance (P>0. 05). Conclusions The high expression of p53 and high level of DI value and SPF value suggest the pathogenesis of pterygium may be out of balance of cell apoptosis and cell proliferation.
