miRNA-495-3p、Hillday交叉识别蛋白在喉癌组织中的表达及其临床意义
Expressions of miRNA-495-3P and Hillday cross-recognition protein in laryngeal carcinoma and their clinical significances
摘要目的:探讨喉癌组织中miRNA-495-3p(miR-495-3p)、Hillday交叉识别蛋白(HJURP)的表达和相关性,及其与患者临床病理特征的关系。方法:选择2023年6月至10月郑州大学第一附属医院30例原发性喉鳞状细胞癌患者的癌组织及相应的癌旁组织(距离癌组织边界1 cm以上)。采用实时荧光定量聚合酶链反应检测miR-495-3p相对表达量;蛋白质印迹法检测HJURP蛋白的相对表达量。采用Pearson法分析miR-495-3p和HJURP在喉癌组织中的相关性。结果:与癌旁组织比较,miR-495-3p在喉癌组织中的相对表达量低(0.28±0.06比1.00±0.00),差异有统计学意义( t=-66.57, P<0.001);喉癌组织中HJURP蛋白的相对表达量高(10.2±5.6比1.0±0.0),差异有统计学意义( t=9.02, P<0.001)。Pearson相关分析结果显示,喉癌组织中miR-495-3p与HJURP呈负相关( r=-0.63, P<0.001)。有淋巴结转移患者miR-495-3p相对表达量低于无淋巴结转移患者(0.23±0.03比0.30±0.05),差异有统计学意义( t= -4.89, P<0.001);HJURP蛋白相对表达量高于无淋巴结转移患者(13.6±5.1比8.8±5.3),差异有统计学意义( t=2.33, P=0.030)。临床分期Ⅲ~Ⅳ期患者miR-495-3p相对表达量低于Ⅰ~Ⅱ期患者(0.23±0.03比0.32±0.04),差异有统计学意义( t=7.22, P<0.001);HJURP蛋白相对表达量高于Ⅰ~Ⅱ期患者(15.3±5.4比6.9±2.2),差异有统计学意义( t=-5.85, P<0.001)。 结论:喉癌中miR-495-3p表达降低,HJURP蛋白表达升高;二者具有相关性。

abstractsObjective:To investigate the expressions and correlations of miRNA-495-3p (miR-495-3p) and Hillday cross-recognition protein (HJURP) in laryngeal carcinoma tissues, and their relationship with clinicopathological features of patients.Methods:Cancerous tissues and corresponding paracancerous tissues (more than 1 cm away from the boundary of cancerous tissue) were selected from 30 patients with primary laryngeal squamous cell carcinoma in the First Affiliated Hospital of Zhengzhou University from June to October 2023. Real-time fluorescence quantitative polymerase chain reaction was used to detect the relative expression of miR-495-3p. The relative expression of HJURP protein was detected by Western blotting. The correlation between miR-495-3p and HJURP in laryngeal carcinoma was analyzed by Pearson method.Results:Compared with paracancerous tissues, the relative expression of miR-495-3p in laryngeal cancer tissues was lower (0.28±0.06 vs. 1.00±0.00), and the difference was statistically significant ( t = -66.57, P < 0.001); the relative expression of HJURP protein in laryngeal carcinoma tissues was higher (10.2±5.6 vs. 1.0±0.0), and the difference was statistically significant ( t = 9.02, P < 0.001). The results of Pearson correlation analysis showed that miR-495-3p was negatively correlated with HJURP in laryngeal cancer tissues ( r = -0.63, P < 0.001). The relative expression of miR-495-3p in patients with lymph node metastasis was lower than that in patients without lymph node metastasis (0.23±0.03 vs. 0.30±0.05), and the difference was statistically significant ( t = -4.89, P < 0.001); the relative expression of HJURP protein was higher than that in patients without lymph node metastasis (13.6±5.1 vs. 8.8±5.3), and the difference was statistically significant ( t = 2.33, P = 0.030). The relative expression of miR-495-3p in patients with clinical stage Ⅲ-Ⅳ was lower than that in patients with stage Ⅰ-Ⅱ (0.23±0.03 vs. 0.32±0.04), and the difference was statistically significant ( t = 7.22, P < 0.001); the relative expression of HJURP protein was higher than that in patients with stage Ⅰ-Ⅱ (15.3±5.4 vs. 6.9±2.2), and the difference was statistically significant ( t = -5.85, P < 0.001). Conclusions:In laryngeal carcinoma, the expression of miR-495-3p is decreased, and the expression of HJURP protein is increased; the two are related.
