食管鳞状细胞癌中热休克蛋白90α和基质金属蛋白酶-2的表达及意义
Expression and clinical significance of HSP 90α and MMP-2 in esophageal squamous cell carcinoma
摘要目的 研究热休克蛋白90α(HSP 90α)和基质金属蛋白酶-2(MMP-2)在食管鳞状细胞癌(简称鳞癌)中的蛋白表达,分析二者表达的相关性及其与临床病理特征之间的关系.方法 采用免疫组织化学Elivision非生物素检测方法,对58例食管鳞癌、30例癌旁正常食管上皮组织切片进行染色,观察、评定HSP 90α和Survivin蛋白表达的阳性率及表达强度.结果 HSP 90α阳性产物主要位于细胞核,在食管鳞癌及食管正常上皮的阳性表达率分别为68.97%和13.33%.MMP-2阳性产物主要位于细胞核,在食管鳞癌及食管断端正常上皮的阳性表达率分别为70.69% 和16.67%.Survivin阳性产物主要位于细胞核,在食管鳞癌及食管断端正常上皮的阳性表达率分别为56.90%和6.67%.在食管鳞癌中,HSP90α表达与MMP-2表达呈正相关(rs=0.534,P=0.000).结论 HSP90α与MMP-2在食管鳞癌的发生、发展中起重要作用.

abstractsObjective To study the expression of HSP 90α and MMP-2 proteins in esophageal squamous cell carcinoma(ESCC),and to analyze the relationships among expression,between protein expression and clinicopathologic factors.Methods Fifty-eight cases of ESCCs,30 cases of pericarcinoma normal tissues were immunohistochemically stained using HSP 90α,MMP-2 and Survivin monoclonal antibodies with ElivisionTM plus non-biotin detection system,the positive expression rate and expression intensity were evaluated.Results The HSP 90α protein positive expression rates were 68.97% and 13.33% in ESCC and normal epithelium respectively that mainly being expressed in nucleus,and the MMP-2 protein positive expression rates were 70.69% and 16.67% in ESCC and normal epithelium respectively that mainly being expressed in nucleus.HSP 90α protein expression was positively correlated with MMP-2.Conclusions HSP 90α and MMP-2 may play important roles in the carcinogenesis and development of ESCC.
