摘要The invention relates to a new method of cloning-expression, which makes it possible to detect and identify proteins capable of binding to the domains phosphorylated by the tyrosine of receptor tyrosine kinases and which uses a new probe for this purpose. This probe includes an amino acid sequence derived from the tyrosine part of the molecular receptor tyrosine, or a functional derivative thereof, it has at least one residue of phosphorylated tyrosine, it is free from the tyrosine kinase part of the receiver and it is detectably marked. The invention also describes a method for preparing this probe, a method for locating on a chromosome a gene encoding a protein capable of binding to the domains tyrosine-phosphorylated by receptor tyrosine kinases, as well as a method used for purifying such a protein by means of said probe. In addition, two proteins are described which have been found to use said cloning process, the proteins GRB-1 and GRB-2, DNA encoding these proteins, as well as methods for detecting these proteins.
